This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Rat IgG2c. Standards or samples are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for Rat IgG2c and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Rat IgG2c, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The OD value is proportional to the concentration of Rat IgG2c. You can calculate the concentration of Rat IgG2c in the samples by comparing the OD of the samples to the standard curve.
IgG2 is mainly used to neutralize antigens or block receptor ligand binding, and its CDC and ADCC effects are very weak, and only EGFR antibodies were marketed based on IGG2 in the early days. However, with the rise of immune checkpoint research, more and more IGG2-based drugs have entered the clinic and entered the market. Although IgG2 has weak binding with C1q, it can still trigger CDC effect when the antigen or antibody solubility is high, and IgG2 is the only subtype that can bind FC-γriia (CD32a). Moreover, this binding can be regulated by single nucleotide polymorphisms (SNPS), which can affect the functional activity of IgG2.
