This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Rat ApoH. Samples (or Standards) and Horseradish Peroxidase (HRP) linked antibody specific for Rat ApoH are added to the micro ELISA plate wells. Rat ApoH in samples (or standards) combines with the coated antibody and HRP linked detection antibody special to Rat ApoH.Excess conjugate and unbound sample or standard are washed from the plate. The substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The OD value is proportional to the concentration of Rat ApoH. The concentration of Rat ApoH in the samples is then determined by comparing the OD of the samples to the standard curve.
Apolipoprotein H (ApoH) is a single-chain plasma protein of about 50 kD. It is a member of the complement control superfamily of proteins. ApoH is also known as beta-2-glycoprotein 1 and activated protein C-binding protein. ApoH has been implicated in a variety of physiologic pathways, including blood coagulation, hemostasis, and the production of antiphospholipid antibodies characteristic of antiphospholipid syndrome. ApoH binds negatively charged phospholipid macromolecules and circulates as a component of lipoprotein particles and as a lipid-free serum protein. ApoH may prevent activation of the intrinsic blood coagulation cascade by binding to phospholipids on the surface of damaged cells. ApoH also inhibits thrombosis by blocking the activation of Coagulation Factor XI but also shows pro-coagulant activity by inhibiting the activation of Protein C. The plasma concentrations of ApoH are strongly associated with metabolic syndrome alterations and vascular disease in type 2 diabetics and could be considered as a clinical marker of cardiovascular risk.
