This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Mouse IgG. Samples (or Standards) and Horseradish Peroxidase (HRP) linked antibody specific for Mouse IgG are added to the micro ELISA plate wells. Mouse IgG in samples (or standards) combines with the coated antibody and HRP linked detection antibody special to Mouse IgG.Excess conjugate and unbound sample or standard are washed from the plate. The substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450±2 nm. The OD value is proportional to the concentration of Mouse IgG. The concentration of Mouse IgG in the samples is then determined by comparing the OD of the samples to the standard curve.
There are five classes of mammalian immunoglobulins:IgA, IgD, IgE, IgM, and IgG. IgG is the most abundant immunoglobulinand is equally distributed in blood and tissue. In mice, the IgG class isfurther divided into four subclasses: IgG1, IgG2a无 IgG2c (strain specific),IgG2b, and IgG3. The general immunoglobulin structure is composed offour polypeptide chains, two heavy and two light chains linkedtogether and to each other by disulfide bonds, creating a tetramericquaternary structure. The resulting tetramer creates two identicalhalves which together form a Y like structure. While the amino-terminalportions that exhibits highly variable amino-acid composition areinvolved in antigen binding, the C terminal constant parts are involvedin complement binding, placental passage and binding to cellmembrane. IgG is involved in response to a foreign antigen. Thepresence of IgG usually signifies a mature antibody response. IgG has amolecular weight of about 150 kDa, it can bind to many pathogensand also plays an important role in antibody-dependent cell-mediatedcytotoxicity. Typically, mouse serum and plasma samples containabout 7 to 10 mg无ml of IgG.
