This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to LAB7-1 . Standards or samples are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for LAB7-1 and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain LAB7-1 , biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The OD value is proportional to the concentration of LAB7-1 . You can calculate the concentration of LAB7-1 in the samples by comparing the OD of the samples to the standard curve.
CD80 is a 60 kD highly glycosylated protein. It is a member of the Ig superfamily and is also known as B7-1,B7,and Ly-53. CD80 is constitutively expressed on dendritic cells and monocytes/macrophages,and inducibly expressed on activated B and T cells. The ligation of CD28 on T cells with CD80 and CD86 (B7-2) on antigen presenting cells (such as dendritic cells,macrophages,and B cells) elicits co-stimulation of T cells resulting in enhanced cell activation,proliferation,and cytokine production. CD80 appears to be expressed later in the immune response than CD86. CD80 can also bind to CD152,also known as CTLA-4,to deliver an inhibitory signal to T cells.
