This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to SAA . Standards or samples are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for SAA and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain SAA , biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The OD value is proportional to the concentration of SAA . You can calculate the concentration of SAA in the samples by comparing the OD of the samples to the standard curve.
SAA is A family of proteins encoded by multiple genes, most of which are synthesized by the liver. SAAl and SAA2 encode acute SAA (A-SAA), SAA3 is a non-functional sequence, and SAA4 encodes a constitutive SAA (C-SAA) in humans. During the acute phase of inflammation, sAA can increase within 24 hours, maintain for 2-3 days, then decrease, and return to normal levels after 7-10 days. It has long been regarded as an acute phase protein, but existing studies suggest that SAA can act as apolipoprotein to promote cholesterol recycling under inflammatory conditions. Macrophages, lymphocytes, endothelial cells and other cells are mobilized to participate in inflammatory response and tissue repair.
