Test Principle
This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to gp130 . Standards or samples are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for gp130 and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain gp130 , biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The OD value is proportional to the concentration of gp130 . You can calculate the concentration of gp130 in the samples by comparing the OD of the samples to the standard curve.
Background
The protein encoded by this gene is a signal transducer shared by many cytokines , including interleukin 6 ( IL6 ) , ciliary neurotrophic factor ( CNTF ) , leukemia inhibitory factor ( LIF ) , and oncostatin M ( OSM ) . This protein functions as a part of the cytokine receptor complex. The activation of this protein is dependent upon the binding of cytokines to their receptors. vIL6 , a protein related to IL6 and encoded by the Kaposi sarcoma-associated herpesvirus , can bypass the interleukin 6 receptor ( IL6R ) and directly activate this protein. Knockout studies in mice suggest that this gene plays a critical role in regulating myocyte apoptosis. Alternatively spliced transcript variants have been described. A related pseudogene has been identified on chromosome 17.
