This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to IFN-γ . Standards or samples are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for IFN-γ and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain IFN-γ , biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 30 nm. The OD value is proportional to the concentration of IFN-γ . You can calculate the concentration of IFN-γ in the samples by comparing the OD of the samples to the standard curve.
Human IFN-γ (interferon-γ) is an important immunomodulatory cytokine belonging to the Type II interferon family. It plays a key role in probiotic and adaptive immune responses, and has a wide range of biological functions, including antiviral, anti-tumor, immunomodulatory, and promoting cell proliferation and differentiation. IFN-γ is mainly produced by activated T cells (such as CD4+ and CD8+ T cells) and natural killer cells (NK cells). It is a non-covalent homologous dimer consisting of two 16 kDa subunits, which is modified by glycosylation to form a biologically active dimer. The molecular weight of IFN-γ is about 16 kDa, but higher molecular weight polymers can be formed under non-denatured conditions. IFN-γ, as an important immunomodulatory cytokine, plays a key role in both natural and adaptive immune responses. It not only has antiviral, anti-tumor and immunomodulatory functions, but also plays an important role in the occurrence and development of autoimmune diseases. Future studies will further reveal the specific mechanisms of IFN-γ in different diseases and provide new ideas for its clinical application.
