This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Human SCCA2. Standards or samples are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for Human SCCA2 and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Human SCCA2, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The OD value is proportional to the concentration of Human SCCA2. You can calculate the concentration of Human SCCA2 in the samples by comparing the OD of the samples to the standard curve.
SCCA2 (squamous cell carcinoma associated antigen 2), also known as SerpinB4, is a serine protease inhibitor encoded by the SERPINB4 gene.It shares up to 92% amino acid sequence similarity with SCCA1 (squamous cell carcinoma associated antigen 1), both of which are produced primarily by different splicing forms of the same gene 1.SCCA2 is widely expressed in normal tissues, especially in squamous epithelial cells, and plays a role in inhibiting apoptosis and promoting cell division 2.Biological activity of SCCA2 SCCA2 belongs to a large family of serine trypsin inhibitors and has the activity of inhibiting a variety of proteases, including chymotrypsin and cathepsin G.This property allows SCCA2 to play an important role in the protection and repair of epithelial cells.Application of SCCA2 in tumor diagnosis SCCA2, as a tumor marker, plays an important role in the diagnosis and management of various squamous cell carcinomas.For cervical cancer in particular, an average of 55% of patients with untreated squamous cell carcinoma of the cervix have positive serum SCCA levels, a proportion that is strongly correlated with clinical stage4.
