This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Human IL18BP. Standards or samples are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for Human IL18BP and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Human IL18BP, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The OD value is proportional to the concentration of Human IL18BP. You can calculate the concentration of Human IL18BP in the samples by comparing the OD of the samples to the standard curve.
Interleukin-18 binding protein is a secreted glycoprotein that fights the activity of IL-18 by binding to IL-18 and blocking its biological activity. Alternative splicing of the IL-18 BP gene produces at least four subtypes with different affinity for IL-18. The IL-18 BP isomers a and c each contain an immunoglobulin (Ig) -like C2-type domain, while the isomers b and d lack a complete IGG-like domain. In mouse models of experimental autoimmune encephalomyelitis (EAE), collagen-induced arthritis (CIA), contact hypersensitivity, lipopolysaccharide-induced shock, and hemophagocytic lymphohistiocytosis (HLH), the anti-inflammatory effects of an interleukin-18 receptor antagonist (IL-18BPa) have been observed in vivo. Marked by a reduction in the development or severity of the disease. IL-18 receptor antagonists prevent the development and progression of atherosclerosis by reducing arterial lipid deposition, immune cell infiltration, and plaque instability. After heart ischemia/reperfusion injury, IL-18 receptor antagonists limit inflammation and improve heart function. In cancer, IL-18 receptor antagonists inhibit tumor progression and metastasis.
