This ELISA kit uses the Competitive-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with Universal FMN. During the reaction, Universal FMN in the sample or standard competes with a fixed amount of Universal FMN on the solid phase supporter for sites on the Biotinylated Detection Ab specific to Universal FMN. Excess conjugate and unbound sample or standard are washed away, and Avidin-Horseradish Peroxidase (HRP) conjugate are added to each micro plate well and incubated. Then a TMB substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns from blue to yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The concentration of Universal FMN in tested samples can be calculated by comparing the OD of the samples to the standard curve.
Flavin mononucleotide, also known as riboflavin-5-phosphate, is a cogroup of the flavoprotein. It plays a vital role in living organisms, especially in electron transfer in biological oxidation processes such as respiration. The structure of the flavin mononucleotide consists of three key components: the isoalloxazine ring (located in the lower left corner and consisting of three thick rings), the ribose chain, and the phosphate group. Although the isoalloxazine ring is similar to the base, it is not a base in the traditional sense, but an important feature of the flavin mononucleotide structure. Instead of the closed-loop structure common in DNA and RNA, ribose strands are open, consisting of four carbon atoms, each attached to a hydroxyl group. Flavin mononucleotides undergo reversible tautosis in a variety of REDOX enzymes, including the oxidizing (FMN), semi-quinone (FMNH•), and reduced (FMNH2) forms. It is a stronger oxidizing agent than nicotinamide adenine dinucleotide and can be involved in the transfer of one or two electrons, so it is particularly useful in this regard. Flavin mononucleotides are widely distributed in organisms and have important effects on biological processes such as energy transfer and REDOX reactions. In addition, it is also used as a food coloring, and its E code is designated as "E101a". The food coloring closely related to flavin mononucleotides, riboflavin 5' -sodium phosphate, can be found in some foods for infants and young children, such as jam, milk, and candy and sugar products.
