Detection Principle Adenosine Triphosphate (ATP) is a high-energy phosphate compound, which is the most direct energy source in the body, and its content is directly related to the state of energy metabolism in the body. Hexokinase (HK) catalyzes the generation of glucose and ATP into glucose-6-phosphate, and glucose-6-phosphate dehydrogenase (G6PD) further catalyzes the dehydrogenation of glucose-6-phosphate into NADPH. NADPH can react with the color developer, and the product has a characteristic absorption peak at 460 nm to determine the ATP content.

The recommended dilution factor for different samples is as follows (for reference only):

Sample Type Dilution Factor
10% Mouse heart tissue homogenate 1
10% Mouse kidney tissue homogenate 1
10% Mouse muscle tissue homogenate 1
10% Mouse liver tissue homogenate 1
10% Mouse brain tissue homogenate 1
10% Fish muscle tissue homogenate 1

The diluent is normal saline (0.9% NaCl) or PBS (0.01 M, pH 7.4). For the dilution of other sample types, please do pretest to confirm the dilution factor.
